چکیده

The relationship between availability and utilization of glucose and production of láclatewas studied in vivoin Walker carcinoma 256, Hepatoma 5123, and Fibrosarcoma 4956 transplanted in rats. The tumors utilized 0.96,0.55, and 0.45 gm of glucose/hr/100 gm wet tumor weight, respectively, corresponding to 28%, 23%, and 32% of the amount supplied. About 35% of the glucose consumed was eliminated as Jáctateregardless of the tumor type. In normoglycemia, the amount of glucose consumed and glycolyzed was directly related to the glucose available which de creased per unit weight as the tumor size increased. During hyperglycemia, produced either by dextrose injections or diabetes, the tumors increased glucose utilization. The increase was maximum during the first few hours; then a "saturation" level

Relationship between oxygen and glucose consumption by transplanted tumors in vivo.

The in vivo consumption of oxygen and glucose was studied in relation to growth in Walker Carcinoma 256, Hepatoma 5123, and Fibrosarcoma 4956 transplanted in rats. Glucose not elim inated as lactate or carbon dioxide in the efferent blood was pre sumed to be retained by the tumor. The retention was so high that growth alone could not account for it. As an alternative, the elimination of glucose...

Modulation of Glucose Transporter 1 (GLUT1) Expression Levels Alters Mouse Mammary Tumor Cell Growth In Vitro and In Vivo

Tumor cells exhibit an altered metabolism characterized by elevated aerobic glycolysis and lactate secretion which is supported by an increase in glucose transport and consumption. We hypothesized that reducing or eliminating the expression of the most prominently expressed glucose transporter(s) would decrease the amount of glucose available to breast cancer cells thereby decreasing their meta...

pH in human tumor xenografts and transplanted rat tumors: effect of insulin, inorganic phosphate, and m-iodobenzylguanidine.

Various strategies to improve the therapeutic index of anticancer agents aim at inducing, by stimulation of aerobic glycolysis, temporary pH differences between malignant and normal tissues which can be exploited to activate cytotoxic agents selectively in tumors. We have investigated whether the pH reduction induced by glucose, the "drug" commonly used to increase lactic acid production in mal...

Tumor-selective modification of cellular microenvironment in vivo: effect of glucose infusion on the pH in normal and malignant rat tissues.

The pH distributions in transplanted neural (TV1A, BT1A) and hepatic (HV1A3) tumors and in brain and kidney of BDIX rats were analyzed as a function of serum glucose concentration (SGC), tumor size, and tissue architecture. Tissue damage during pH measurements in vivo could be minimized by the use of pH microelectrodes with tip diameters of less than or equal to 10 micrometers. In normoglycemic...

A review of the health effects of green tea catechins in in vivo animal models.

There is good evidence from in vitro studies that green tea catechins have a role in protection against degenerative diseases. However, the concentrations used in vitro are often higher than those found in animal or human plasma, and so in vivo evidence is required to demonstrate any protective effect of catechins. This article summarizes the most interesting in vivo animal studies on the prote...
